چکیده

In a previous publication from this laboratory (1), radioactive diisopropylfluorophosphate (DFP32), a potent and irreversible esterase inhibitor, was shown to be a suitable label for granulocytes. Evidence was presented that the label is firmly attached to granulocytes; it does not modify the viability of the cells, and the label is not reutilized when the cells are degraded. Experiments designed to determine whether or not elution of the label occurs failed to provide such evidence. More recent studies (2), carried out with tritiated DFP and autoradiography, have confirmed the observation that the label is attached to granulocytes and little, or not at all, to lymphocytes or to monocytes. A method for labeling granulocytes in vivo with DFP32 was described in the previous publication (1). The purpose of this paper is to describe a method for labeling granulocytes in vitro, and to describe the manner in which granulocytes so labeled leave the circulation of normal subjects. Several preliminary reports of this work have been published (3, 4).

The Kinetics of Granulopoiesis in Normal Man.

I N 1955, when studies were initiated in our laboratory to determine the suitability of radioactive diisopropylfluorophosphate ( DFP32) as a label for granulocytes, information concerning the rate of production and movement of these cells was very meager as compared with knowledge of erythrokinetics. Since that time, with the development and application of the DFP32 cytoplasmic granulocyte labe...

Leukokinetic studies. XIV. Blood neutrophil kinetics in chronic, steady-state neutropenia.

The kinetics of blood neutrophils was investigated by means of the in vitro radioactive diisopropyl fluorophosphate method in 35 patients with a chronic, steady-state neutropenia. There were 17 patients in whom the half disappearance time of neutrophils was normal. In 10 of these patients, the production of neutrophils was low and in 7, production was normal. In 18 patients the half disappearan...

Leukokinetic Studies. Ix. Experimental Evaluation of a Model of Granulopoiesis.

Kinetic models of granulopoiesis have been constructed from data obtained by labeling neutrophilic granulocytes with tritiated thymidine (1, 2), with radioactive phosphorus (3), and with diisopropyl fluorophosphate (DFP32) (4). In many respects these models are remarkably similar in their portrayal of the concatenated compartments of normal granulopoiesis. However, certain questions, particular...

Spatiotemporal patterns of GFAP upregulation in rat brain following acute intoxication with diisopropylfluorophosphate (DFP).

We previously demonstrated that the organophosphate diisopropylfluorophosphate (DFP) induced region-specific delayed neuronal injury in rat brain. The goal of this study was to examine the spatiotemporal pattern of the astroglial response after acute intoxication of rats with DFP. Rats were euthanized at varying times between 1 h to 7 days after DFP administration (9mg/kg, i.p.). The distributi...

Platelet tagging with tritium labeled diisopropylfluorophosphate.

By EDWARD ADELSON, RICHARD M. KAUFMAN, Cmo BERDEGUEZ, ARNOLD A. LEAR AND JACK J. RHEINGOLD F OR THE PAST 10 years, P32 labeled diisopropylfluorophosphate has been used to tag platelets.”2 This compound attaches to various proteins, during which process it changes from diisopropylfiuorophosphate ( DFP) to diisopropylphosphate (DIP).3 The DIP is irreversibly bound to the protein molecule until th...
